Yes you keep all your ability points and abilities unlocked. All quests should be available to complete again in NG+, including the side quests. Justput your sleep darts in the tool slot next to your shield. Once equipped, hold down left on the D-pad to equip the sleep darts, then just aim and fire them like you would your bow.
